These non-coding RNA-activators (ncRNA-a) have a crucial role in turning genes on and off during early embryonic development, researchers say, and have also been connected with diseases, including some cancers, in adults.

In an online article of the journal Nature, a team of scientists led by Wistar's Ramin Shiekhattar, Ph.D., detail the mechanism by which long non-coding RNA-activators promote gene expression. They show how these RNA molecules help proteins in the cell to create a loop of DNA in order to open up genes for transcription. Their experiments have also described how particular ncRNA-a molecules are related to FG syndrome, a genetic disease linked to severe neurological and physical deficits. "These ncRNA-activators can activate specific genes by working with large protein complexes, filling in a big piece of the puzzle," said Shiekhattar, Herbert Kean, M.D., Family Professor and senior author of the study. "Our DNA encodes thousands of these ncRNA-activators, each with a role in timing the expression of a specific gene. As we learn more about non-coding RNA, I believe we will have a profoundly better understanding of how our genes function."

Their findings also provide a plausible mechanism of how locations along chromosomes, classically known as "enhancer" elements, can influence the expression ("reading") of genes located 5,000 to 100,000 base pairs ("letters") of DNA away. According to their findings, ncRNA-a molecules bind to large protein complexes to form a loop of DNA, which then opens up the gene to the molecular machinery that transcribes DNA. "There is an abundance of evidence to indicate that enhancers are critical components of transcription during embryonic development and disease process," Shiekhattar said.

"Non-coding RNAs are probably one of the earliest molecules that determine spatial and temporal gene expression in a developing embryo," Shiekhattar said. "These enhancers can help turn genes on and off as a growing embryo would need, but as we have seen in other genetic mechanisms of embryonic development, they can lead to cancer if they are switched on inappropriately in adult cells."

In the classic "central dogma" of biology, chromosomal DNA is transcribed into RNA, which is then translated by the cell into proteins. In recent years, however, scientists have found that not all transcribed RNA molecules become translated into proteins. In fact, studies have shown that large portions of the genome are transcribed into RNA that serve tasks other than functioning as blueprints for proteins. In 2010, the Shiekhattar lab first published the discovery of these ncRNA enhancer molecules in the journal Cell(2010 Oct 1;143(1):46-58), and theorized on their role as "enhancers" of gene expression. Since then, laboratories around the world have published and linked ncRNAs not only to transcriptional enhancers but also to certain diseases, including some cancers.

To discover how such enhancer-like RNAs function, the Shiekhatter laboratory deleted candidate molecules with known roles in activating gene expression, and assessed if they were related to RNA-dependent activation. They found that depleting components of the protein complex known as Mediator specifically and potently diminished the ability of ncRNA-a to start the process of transcribing a gene into RNA. Further, they found that these activating ncRNAs can attach to Mediator at multiple locations within the Mediator protein complex, and Mediator itself can interact with the enhancer element site on DNA that encodes these activating ncRNAs. Their results also determined how mutations in a protein that makes up the Mediator complex, called MED12, drastically diminishes Mediator's ability to associate with activating ncRNAs.

Mutations in the MED12 protein are a marker for FG syndrome (also know as Opitz?Kaveggia syndrome), a rare genetic disorder that leads to abnormalities throughout the body and varying degrees of physical and neurological problems. "This clearly shows how activating ncRNAs can influence disease development, an idea that has been gaining evidence in the scientific literature," Shiekhattar said. To confirm that ncRNA-a works with Mediator to form a loop in DNA, the researchers used a technique called chromosome conformation capture (3C) to gain a better understanding of the three-dimensional structure of chromosomes. Their results show how Mediator gets a foothold of sorts on the portion of DNA that encodes the ncRNA-a, and twists the DNA to form a loop.

"The looping mechanism serves to physically bring together a distant enhancer element with the start site of the targeted gene, allowing Mediator to recruit the proteins responsible for reading the gene to the location," Shiekhattar said. "It is at least one answer to how these classical enhancer elements function while being physically distant from their target genes."

Two other noteworthy extras are the automatic duplexer (for printing on both sides of a page) and the ability to print on printable optical discs, although you may have to check the onscreen manual to figure out where the disc printing options are hidden in the software that ships with the printer.

Setup and Speed
Setting up the iP7220 was standard fare. For my tests, I connected it by USB cable to a system running Windows Vista. The speed for business applications is best described as a little slow but within a tolerable range for a home or home office printer. On our business applications suite, I timed it (using QualityLogic's hardware and software for timing) at an effective 2.5 pages per minute (ppm), making it only about 70 percent as fast as the Officejet 6100 ePrinter, at 3.6 ppm, and even slower compared with the slightly more expensive Editors' Choice Epson WorkForce 60, at 4.1 ppm.

Making up somewhat for the speed for business applications is the printer's photo speed, which averages a reasonably fast 1 minute 5 seconds for a 4 by 6. That makes it essentially tied with the Officejet 6100 for photo speed, and about twice as fast as the WorkForce 60.

Output Quality
Also helping to make up for the slow speed for business applications is that the output quality is above par overall, with better text and photo quality than most inkjets and par quality graphics. The text isn't quite a match for a typical laser, and I wouldn't use it for, say, a resume. However, its far more readable at small font sizes that most ink jets can manage, and it's good enough for almost any business use.

Graphics quality is dead on par for an inkjet, which makes it easily good enough for any internal business use, including PowerPoint handouts. As with many printers, the iP7220 has a tendency to lose thin lines against a solid dark background, but depending on how critical an eye you have, you may consider the quality good enough for output going to an important client or customer. Photo quality is in the top tier for inkjets that aren't aimed specifically at photo enthusiasts or professionals, which makes them noticeably better than the photos you'll get from many drug stores.

Sony PlayStation 4 to stream PS3 titles


Sony intends to offer technology that will allow users to stream PlayStation 3 games on its forthcoming PlayStation 4 console, according to the Wall Street Journal. This latest speculation surrounding the next generation of the popular video game console comes ahead of its expected revelation in New York later this week.

According to the report, "people familiar with the company?s plans" have said that the new system will enable users to play games delivered over the Internet and is designed to use current PlayStation 3 titles, in addition to the ability to play new games stored on standard optical discs. The report also points out that the move doesn?t come as much of a surprise since Sony acquired Gaikai, a company that used to offer light-weight streaming games, last July.

The introduction of streaming titles could prove to be very significant, potentially ushering in the introduction and rise of cloud gaming, giving gamers direct access to a wide range of titles across devices. The system will also make it possible for Sony to introduce its own online game rental service.

MISHAWAKA ? In a meet in which it was hard to stand out, South Bend St. Joseph senior Trevor Carroll showed how ready he is to head back to the IHSAA State Championships next week.

Carroll left the Penn pool with three firsts, including the distinction as the only double individual winner from the Penn Sectional Saturday.

Carroll will return to defend his state title in the 200 freestyle after winning that event Saturday before he turned in virtuoso performances in his other two victories.

Penn took the team title, its fourth straight, with 521 points. St. Joseph finished second with 447.

The sectional field proved to be the deepest, at least since the middle of the last decade, as seven sectional and five pool records were set. Also, there were multiple qualifiers in five events.

?This is the fastest the sectional has been in my career,? Carroll said. ?It was fun to be a part of this.?

Carroll opened the day with a 1:41.03 clocking to win the 200 free. While an outstanding time, it was over two seconds slower than his sectional record time of a year ago.

Carroll, however, won the 500 free in an outstanding 4:27.55 effort. That shattered the sectional record by more than 13 seconds and broke the pool record (4:31.25) set in 1998.

?It just felt awesome,? said Carroll. ?I felt most of the season I wanted to swim this event, but I wanted to do what was best for the team first. Our sprinters came along great, so that opened up the 500 for me.?

He brought the rest of the field along as well, as the next three finishers (in order, Penn?s Bowen Anderson, Riley?s James Salmon and Penn?s Hudson Bursch) all beat the state qualifying time.

After helping St. Joe to second in the 200 free relay (in which the Indians qualified, as well), Carroll finished the day by anchoring the 400 free relay.

Penn had a solid lead through the first three legs of the race, leaving its anchor, Seamus Lucero-Dixon, with a body-length lead. Carroll, however, made that up during the first 50 yards, then brought home a sectional and pool record time of 3:09.87. Billy Miller, Anthony Kolbus and Andre Marin swam the first three legs.

?When I anchor relays,? Carroll explained, ?I don?t look where anyone else is at during the first half. I just swim my race. After that, I just wanted to hold off (Lucero-Dixon) and we got a school record out of it, too.?

Penn also qualified with a 3:11.44 clocking, capping a banner day for the Kingsmen.

Penn opened the meet with a sectional and pool record in the 200 medley relay as the Anderson brothers (Tanner and Bowen) joined Mate Kovacs and Lucero-Dixon to win in 1:34.65.

Tanner Anderson came back to win the 200 IM (1:55.39) before Kovacs took the 50 free (:21.86) to give the Kingsmen firm control in the team standings.

From there Penn came through with four firsts. Bowen Anderson (a sophomore) won the 100 butterfly in a sectional and pool record time of :50.37, followed by Lucero-Dixon, who took care of the 100 free (:47.63). The 200 free relay of Tanner Anderson, Kovacs, Travis Bavar and Lucero-Dixon set Penn?s final sectional and pool record of the day with a 1:25.96 time before freshman Kyle Morris won the 100 backstroke in :54.34.

?We?re right on track for next week,? Penn coach Jess Preston said. ?We kept Seamus and Mate to just one individual event so they could help us in the relays and Tanner and Bowen were outstanding for us. We wanted to get them through in four events and they got it done.?

The final new sectional mark came from St. Joe?s Chris Bals, who took the 100 breaststroke in :57.36.

G20 defuses talk of "currency war", no accord on debt

MOSCOW (Reuters) - The Group of 20 nations declared on Saturday there would be no 'currency war' and deferred plans to set new debt-cutting targets in an indication of concern about the fragile state of the world economy.

Japan's expansive policies, which have driven down the yen, escaped criticism in a statement thrashed out in Moscow by financial policymakers from the G20, which groups developed and emerging markets and accounts for 90 percent of the world economy.

After late night talks, finance ministers and central bankers agreed on wording closer than expected to a joint statement issued last Tuesday by the Group of Seven rich nations backing market-determined exchange rates.

A draft communique seen by delegates on Friday had steered clear of the G7's call for economic policy not to be targeted at exchange rates. But the final version included a G20 commitment to refrain from competitive devaluations and stated monetary policy would be directed at price stability and growth.

"The language has been strengthened since our discussions last night," Canadian Finance Minister Jim Flaherty told reporters. "It's stronger than it was, but it was quite clear last night that everyone around the table wants to avoid any sort of currency disputes."

The communique did not single out Japan for aggressive monetary and fiscal policies that have seen the yen drop 20 percent, a trend that may now continue.

"The market will take the G20 statement as an approval for what it has been doing -- selling of the yen," said Neil Mellor, currency strategist at Bank of New York Mellon in London. "No censure of Japan means they will be off to the money printing presses."

The statement reflected a substantial, but not complete, endorsement of Tuesday's statement by the G7 nations - the United States, Japan, Britain, Canada, France, Germany and Italy.

"We all agreed on the fact that we refuse to enter any currency war," French Finance Minister Pierre Moscovici told reporters.

NO FISCAL TARGETS

The text also contained a commitment to credible medium-term fiscal strategy, but stopped short of setting specific goals.

A debt-cutting pact struck in Toronto in 2010 will expire this year if leaders fail to agree to extend it at a G20 summit of leaders in St Petersburg in September.

European Economic and Monetary Affairs Commissioner Olli Rehn said he expected concrete debt targets to be agreed at the September meeting.

"We have a common view on the need to have a credible medium-term plans for fiscal consolidation, which is also essential so we have foundation for sustainable growth," he told Reuters.

The United States says it is on track to meet its Toronto pledge but argues that the pace of future fiscal consolidation must not snuff out demand. Germany and others are pressing for another round of binding debt-cutting goals.

Backing in the communique for the use of domestic monetary policy to support economic recovery reflected the U.S. Federal Reserve's commitment to monetary stimulus through quantitative easing, or QE, to promote recovery and jobs.

QE entails large-scale bond buying -- $85 billion a month in the Fed's case -- that helps economic growth but creates money, much of which has leaked into emerging markets, threatening to destabilize them.

That was offset in the communique by a commitment to minimize "negative spillovers" of the resulting financial flows that emerging markets fear may pump up asset bubbles and ruin their export competitiveness.

"Major developed nations (should) pay attention to their monetary policy spillover," Vice Finance Minister Zhu Guangyao was quoted by state news agency Xinhua as saying in Moscow.

"Major developed countries' implementation of excessively relaxed currency policy has an influence on the world economy."

Russia, this year's chair of the G20, said the group had failed to reach agreement on medium-term budget deficit levels and also expressed concern about ultra-loose policies that it and other big emerging economies say could store up trouble for later.

Finance Minister Anton Siluanov said a rebalancing of global growth required more than an adjustment of exchange rates.

"Structural reforms in all countries, either with a positive or negative balance of payments, should play a bigger role," he said in an address to Saturday's talks.

The G20 put together a huge financial backstop to halt a market meltdown in 2009 but has failed to reach those heights since. At successive meetings, Germany has pressed the United States and others to do more to tackle their debts. Washington in turn has urged Berlin to do more to increase demand.

On currencies, the G20 text reiterated its commitment last November, "to move more rapidly toward mores market-determined exchange rate systems and exchange rate flexibility to reflect underlying fundamentals, and avoid persistent exchange rate misalignments".

It said disorderly exchange rate movements and excess volatility in financial flows could harm economic and financial stability.

Microsoft, on the other hand, continues to praise its new operating system, claiming that the 60 million sold copies are in line with those recorded by Windows 7 more than three years ago.

Valerie Walkowiak, medical integration coordinator at the Loyola Center for Fitness, gives some ideas for how to keep up your fitness routine until you feel it?s safe to go back to the gym.

?Weather permitting, it?s always great to just get out of the house and walk or run to get in some cardio exercise. Just make sure you wear the proper clothes to keep warm. This includes layers of clothing, a hat, scarf, gloves and appropriate shoes,? Walkowiak said.

If you don?t have exercise equipment in the house and the weather makes it impossible to exercise outside she suggests climbing up and down a staircase, jogging in place or performing jumping jacks.

If you?re looking for a little more, she suggests creating your own circuit training exercises. This includes 2-3 minutes of cardio exercise alternated by 30-60 seconds of strength-training exercise to get your heart rate up.
